Optimization of Crude Protease Production from Bacillus thuringiensis HSFI-12 and Thrombolytic Activity Its Enzyme Dialysate

Cardiovascular disease (CVD) is among the leading causes of death in world caused by thrombosis. Thrombosis formation excessive blood clots on walls vessels called thrombus. This could lead to fatal blockage heart muscle or brain. can be treated using enzyme-type drugs such as thrombolytic proteases. There have been many studies bacteria producing enzymes isolated from fermented foodstuffs. However, commercial production bacterial fulfill need agents still limited, causing high price CVD therapy. Bacillus sp. HSFI-12 (Holothuria scabra Fermented Intestine-12) intestine sea cucumber Holothuria had reported a competitive activity Nattokinase. study aimed determine optimum incubation time based enzyme after bacterium molecular identification was done. It also obtain dialysate crude protease and then obtained dialysate. Thrombolytic tested 4 different types (A, B, AB O). Results showed that strain shared similarity level 99.80 %, with thuringiensis. Activity incubated cultures peaked at 48 h 191.5 U/mL The concentrated precipitation dialysis process (dialysate) 2-times higher 355,7 U/mL. percentage lysis produced groups A, O range values of 66.423 - 67.656 % while those produce are 77.564 78.861 %.
